When applied to the skin and penetrates, a compound may be prone to metabolism that will convert it to an inert, more potent or toxic substance. It can also change its distribution and excretion profile. While skin metabolisms can be determined using cultured human cells, skin homogenate or microsomes or full thickness human skin, no method was officially validated. ESAC estimates that a method will not be validated before 2012. This course will review the skin as a metabolic organ, describe regulatory status and look into optional testing.
Module 1: Definitions:
• Toxicokinetics
• Skin metabolism
• Is the skin a metabolic organ?
• Passing first pass effect
Regulatory status
• ECVAM- ESAC position
• ICCVAM position
Module 2:
• Skin penetration
• Testing for systemic metabolism
Module 3: In Vitro Methods:
